GENEVA (AP) ? Swiss pharmaceutical company Roche Group raised its offer for Illumina Inc. to $51 cash per share Thursday, in a bid to speed up a takeover of the U.S. diagnostics company.

The offer represents an increase of almost 15 percent on Roche's previous offer of $44.50 per share that was unanimously rejected by Illumina's board.

"Based on our discussions with Illumina shareholders we have seen interest to accelerate the takeover process," said Roche CEO Severin Schwan in a statement.

But Illumina immediately cautioned its shareholders to wait before accepting the new offer, which values the San Diego-based DNA sequencing specialist at about $6.5 billion.

"Illumina's board of directors will thoroughly review Roche's revised proposal and make a recommendation to stockholders regarding the proposal in due course," it said. The offer runs until April 20.

Analysts had expected Roche to increase its bid after Illumina's share price rose to about $50 following the initial takeover offer.

"We see a win-win situation for both sides in a takeover," Zuercher Kantonalbank said in an analyst note.

Roche shares were down 1.1 percent at 158.10 Swiss francs ($174.93) on the Zurich exchange in morning trading.

PHOENIX (AP) ? It's been a year since the Fiesta Bowl scandal embarrassed legislators, but at least some Arizona lawmakers still like their perks.

The Arizona Diamondbacks have again offered free tickets to legislators for the baseball team's first game of the season, and a team executive said Tuesday that at least 40 of the 90 senators and representatives have accepted so far.

"A good time," said Rep. Doris Goodale, R-Kingman, said when asked what she she'll get out of going to the game April 6.

Lawmakers won't break any rules by accepting the passes. And Sen. Judy Burges, R-Sun City West, said the tickets she's accepting for herself and her husband provide an opportunity for lawmakers to socialize and get to know each other away from the pressure-cooker of the Capitol.

The Fiesta Bowl scandal has led to criminal charges alleging bowl personnel were illegally reimbursed for campaign contributions. The politicians involved apparently weren't aware of the scheme.

In a separate issue that directly touched legislators, the Fiesta Bowl's internal investigation found that numerous lawmakers accepted free game tickets and trips to college football games. Many had not reported them on financial disclosure forms.

A prosecutor who investigated whether legislators violated gift and disclosure laws concluded no prosecution was warranted because state laws are so complex and contradictory.

The Diamondbacks' giveaway doesn't violate the state's general ban on giving gifts to lawmakers because the offer is made to all 90 members of the Legislature.

Team President Derrick Hall said the free tickets provided legislators are part of a tradition that he started about six years ago to make opening day a community celebration.

"It's just focused on opening day, making that more of a special event," Hall said. He also said the team makes sure it complies with all applicable laws.

The team's website says tickets for opening day range from $5 to $160, but Hall said the tickets that legislators will get will likely be unsold ones for lower-price seats in Chase Field's upper deck.

Hall said the team doesn't consider the event as a lobbying move, and team lobbyist Tom Dorn said the only legislative issue that he has worked on for the team so far this session has been a still-pending bill to allow guns in public buildings.

The legislation has been amended to exclude publicly-owned venues such as the sports stadium used by the baseball team.

"We simply want to protect our fans," Dorn said.

Burges said she considered the Fiesta Bowl scandal when deciding whether accepting the Diamondbacks' offer would be an image problem for the Legislature.

She said she concluded that the Fiesta Bowl's offerings made to other lawmakers were in a different league. "Those tickets to the football games were very expensive," she said.

Goodale said there are no strings attached to accepting the seats that she described as "up in the rafters."

"I never, ever take anything with any expectation there's a return. That's clear," Goodale said.

Hall said the team wasn't providing lawmakers with anything at the game, but Dorn said his lobbying firm, which has other clients besides the Diamondbacks, separately had invited lawmakers to a pregame gathering at a nearby restaurant.

Rep. Debbie McCune Davis, D-Phoenix, said she hadn't decided yet whether to accept opening day tickets, but she said she'd pay for them if she does, as she said she'd done previously.

McCune Davis said the real issue is that legislation proposed in the wake of the Fiesta Bowl scandal to tighten the state's gift and disclosure requirements remains bottled up in the Legislature, not getting hearings.

"I think the public expects us to take action," she said.

Brain insulin resistance contributes to cognitive decline in Alzheimer's disease

Insulin resistance in the brain precedes and contributes to cognitive decline above and beyond other known causes of Alzheimer's disease, according to a new study by researchers from the Perelman School of Medicine at the University of Pennsylvania. Insulin is an important hormone in many bodily functions, including the health of brain cells. The team identified extensive abnormalities in the activity of two major signaling pathways for insulin and insulin-like growth factor in non-diabetic people with Alzheimer's disease. These pathways could be targeted with new or existing medicines to potentially help resensitize the brain to insulin and possibly slow down or even improve cognitive decline.

This is the first study to directly demonstrate that insulin resistance occurs in the brains of people with Alzheimer's disease. The study is now online in the Journal of Clinical Investigation.

"Our research clearly shows that the brain's ability to respond to insulin, which is important for normal brain function, is going offline at some point. Insulin in the brain not only modulates glucose uptake, but also promotes the health of brain cells ? their growth, survival, remodeling, and normal functioning. We believe that brain insulin resistance may be an important contributor to the cognitive decline associated with Alzheimer's disease," said senior author, Steven E. Arnold, MD, professor of Psychiatry and Neurology. Arnold is also the director of the Penn Memory Center, a National Institute on Aging-designated Alzheimer's Disease Core Center. "If we can prevent brain insulin resistance from occurring, or re-sensitize brain cells to insulin with any of the currently available insulin-sensitizing diabetes medicines, we may be able to slow down, prevent, or perhaps even improve cognitive decline.

The risk of developing Alzheimer's disease is increased by 50 percent in people with diabetes. Type 2 diabetes is due to insulin resistance and accounts for 90 percent of all diabetes. The defining clinical feature of Type 2 diabetes (and Type 1 "juvenile" diabetes) is hyperglycemia - high levels of sugar in the blood ? but there is no evidence that the brain in Alzheimer's is hyperglycemic. Insulin acts differently in the brain than in the rest of the body. Researchers found that insulin resistance of the brain occurs in Alzheimer's disease independent of whether someone has diabetes, by excluding people with a history of diabetes from this study.

The investigators used samples of postmortem brain tissue from non-diabetics who had died with Alzheimer's disease, stimulated the tissue with insulin, and measured how much the insulin activated various proteins in the insulin-signaling pathways. There was less insulin activation in Alzheimer's cases than in tissue from people who had died without brain disease. Other proteins linked to insulin action in the brain were abnormal in Alzheimer's disease samples. These abnormalities were highly correlated with episodic memory and other cognitive disabilities in the Alzheimer's disease patients.

In tissue from people with Alzheimer's disease and mild cognitive impairment (MCI), researchers found that changes to a protein called insulin receptor substrate-1 (IRS-1 pS636/639 and pS616) in brain cells were linked to the severity of memory impairments regardless of age, sex, diabetes history, or apolipoprotein E (APOE) gene status. Levels of IRS-1 were also significantly associated with, but not likely to affect, the presence of amyloid beta plaques and neurofibrillary tangles, the signature markers of Alzheimer's disease. This suggests that insulin resistance contributes to cognitive decline independent of the classical pathology of Alzheimer's disease.

Researchers noted that three insulin-sensitizing medicines are already approved by the FDA for treatment of diabetes. These drugs readily cross the blood-brain barrier and may have therapeutic potential to correct insulin resistance in Alzheimer's disease and MCI. "Clinical trials would need to be conducted to determine the impact the drugs have on Alzheimer's disease and MCI in non-diabetic patients," said Dr. Arnold.

Question: A friend and I purchased a tour from Friendly Planet to Ecuador last July. We booked a round-trip flight on American Airlines from Dallas to Miami to arrive in time to connect to the LAN flight to Quito, Ecuador.

We purchased flight insurance with Access America Insurance for the super-saver flights from Dallas to Miami. In February I received an email from Friendly Planet saying that the tour had been canceled due to too few participants. I was given the choice to receive a refund or to schedule another tour later in the year. I decided to reschedule.

However, Access America denied the claim I made to cover the costs of changing the American Airlines flights, which was $137 for each of us.

I had spoken to two representatives from Access America explaining what had occurred, and both encouraged me to file the claim since it was not my fault that the trip was canceled. The process was time-consuming.

I have written a letter to Access America asking for a second review. I think Access America insurance is bogus at best since the trip was interrupted because it was never started. What is the purpose of insurance if not to cover the unexpected? ? Catherine Markland, Whitney, Texas

Answer: If your insurance policy covered a canceled tour, then Access America should have paid up.

But did it? I checked with the insurance company, and rescheduling a trip because of a lack of participation in a tour isn?t covered. I agree with you that when you buy insurance, you?re left with the impression that you would be protected if something goes wrong on your vacation. But you have to read the fine print.

Most travel insurance policies cover specific situations described in their terms. There are a number of common events, such as illness or injury to an insured, a traveling companion or close family member, plus a list of other health, transportation, accommodation, political and work circumstances, that are covered reasons for canceling or interrupting a trip.

It appears you purchased your policy to cover your airline ticket, through your airline?s site. I?m a little dubious of the insurance offered through airlines, because they can be extremely restrictive. (I?ve heard insurance insiders joke that you have to die in a plane crash or lose a limb in order to make a successful claim, but I?m sure that?s an exaggeration.)

Insurance purchased through an airline is often an afterthought ? and optional purchase with your airline ticket. Travel insurance should never be an afterthought. You?re much better off shopping around for the right policy than clicking a button when you?re booking a ticket and then assuming you?ll be covered.

I contacted Access America on your behalf. It reviewed your case and ruled that your tour cancellation wasn?t covered. I?m sorry.

Update: Some of you will recognize this story ? it?s based on the very first ?case dismissed? post I wrote last year. About a month after I covered Markland?s unsuccessful claim, Friendly Planet decided to reimburse her for her flight, after all.

Besieged gunman boasted he brought France to its knees

TOULOUSE, France (Reuters) - A besieged gunman suspected of shooting dead seven people in the name of al Qaeda boasted to police on Wednesday he had brought France to its knees and said his only regret was not having been able to carry out his plans for more killings.

In an unfolding drama that has riveted France, about 300 police, some in body armor, cordoned off a five-storey building in a suburb of Toulouse where the 24-year-old Muslim shooter, identified as Mohamed Merah, is holed up.

Authorities said the gunman, a French citizen of Algerian origin, had been to Pakistan and Afghanistan, where he claimed to have received training from al Qaeda.

Merah told police negotiators he had killed three French soldiers last week and four people at a Jewish school in Toulouse on Monday to avenge the deaths of Palestinian children and because of the French army's involvement in Afghanistan.

"He has no regrets, except not having more time to kill more people and he boasts that he has brought France to its knees," Paris Prosecutor Francois Molins, part of the anti-terrorist unit leading the investigation, told a news conference.

The gunman, who filmed his killings with a small camera, had already identified another soldier and two police officers he wished to kill, Molins said. The gunman had repeated promises to surrender this evening to members of the elite RAID unit surrounding the house, which had been evacuated of its other residents.

"He has explained that he is not suicidal, that he does not have the soul of a martyr and that he prefers to kill but to stay alive himself," Molins said.

Sarkozy, who is running for re-election in five weeks time, paid tribute at a ceremony in an army barracks in Montauban, near Toulouse, to the three soldiers of North African origin killed last week. A fourth soldier of Caribbean origin is in a coma.

"Our soldiers have not died in the way for which they had prepared themselves. This was not a death on the battlefield but a terrorist execution," Sarkozy said, standing before three coffins draped in the French flag after paying his respects to bereaved relatives.

"We must remain united. We should in no way yield to discrimination or vengeance," he said in his eulogy. "France can only be great in unity. We owe it to the memory of these men, we owe it to the three murdered children, to all the victims."

Sarkozy's appeal for national unity came after far-right leader Marine Le Pen, a rival presidential candidate, said France should wage war on Islamic fundamentalism.

Interior Minister Claude Gueant said Merah was a member of an ideological Islamic group in France but this organization was not involved in plotting any violence.

He said Merah had thrown a Colt 45 pistol of the kind used in all the shootings out of a window of the block of flats, where he has been living, in exchange for a mobile phone, but was still armed.

Two police officers were injured in a firefight with the gunman after police swooped at 3 a.m. local time (0200 GMT).

Police sources said they had conducted a controlled explosion of the suspect's car at around 9:00 a.m. (0800 GMT) after discovering it was loaded with weapons. Officials said police had also arrested Merah's girlfriend and his brother, who is also known to authorities as a radical Islamist.

Gueant said Merah had contacted the first soldier he attacked on the pretext of wanting to buy his motorcycle.

Investigators identified the IP address he used - that of his mother - because he was already under surveillance for radical Islamist beliefs.

"We knew, and that is why he was under surveillance, that he had travelled to Afghanistan and Pakistan," the minister said.

Merah's telephone was tapped from Monday and with the help of other information the police decided to raid his house. Merah has a criminal record in France, Gueant said, but nothing indicating such an attack was possible.

A police source told Reuters that investigators had also received a tipoff from a scooter repair shop in Toulouse where the gunman asked to change the color of the Yamaha scooter used to flee the shootings and to remove a GPS tracker device.

A group of young men from Merah's neighborhood described him as a polite man of slight build who liked football and motorbikes and did not seem particularly religious.

"He isn't the big bearded guy that you can imagine, you know the cliche," said Kamal, who declined to give his family name. "When you know a person well you just can't believe they could have done something like this."

Sarkozy had been informed of the standoff early in the morning, officials said. The president's handling of the crisis could be a decisive factor in determining how the French people vote in the two-round presidential elections in April and May.

The Jewish victims from the Ozar Hatorah school were buried in Jerusalem on Wednesday. Parliament speaker Reuben Rivlin said in his eulogy at the hill-top cemetery that the attack was inspired by "wild animals with hatred in their hearts".

Authorities said on Tuesday that the gunman had apparently filmed his rampage through the school with a camera strapped to his body. He wounded Rabbi Jonathan Sandler as he entered the building, then shot an 8-year-old girl in the head, before returning to kill Sandler and his two children, who had rushed to his side, at point blank range.

Immigrants and Islam have been major themes of the campaign after Sarkozy tried to win over the voters of Le Pen, who accused the government on Wednesday of underestimating the threat from fundamentalism.

"We must now wage this war against these fundamentalist political and religious groups that are killing our children, that are killing our Christian children, our Christian young men, young Muslim men and Jewish children," she told the i-Tele news channel, questioning the decision to deploy in Afghanistan.

But leaders of the Jewish and Muslim communities said the gunman was a lone extremist.

France's military presence in Afghanistan has divided the two main candidates in the election. Socialist frontrunner Francois Hollande has said he will pull them out by the end of this year while Sarkozy aims for the end of 2013.

SANFORD, Fla. (AP) ? After declaring victories in getting federal and state officials to investigate the case of an unarmed black teenager shot to death by a neighborhood watch captain, civil rights leaders continued to pressure authorities to make an arrest.

At a town hall meeting Tuesday evening in Sanford, Fla., where the fatal shooting of 17-year-old Trayvon Martin took place last month, officials from the NAACP, the American Civil Liberties Union and the Nation of Islam urged residents to remain calm but demand that the shooter, George Zimmerman, be arrested.

Zimmerman has not been charged in the Feb. 26 shooting and has said he shot Martin, who was returning to a gated community in the city after buying candy at a convenience store, in self-defense after Martin attacked him. Police said Zimmerman is white; his family says he is Hispanic.

"I stand here as a son, father, uncle who is tired of being scared for our boys," said Benjamin Jealous, national president of the NAACP. "I'm tired of telling our young men how they can't dress, where they can't go and how they can't behave."

The case has ignited a furor against the police department of this Orlando suburb of 53,500 people, prompting rallies and a protest in Gov. Rick Scott's office Tuesday. The Justice Department's Civil Rights Division said it is sending its community relations service this week to Sanford to "address tension in the community."

Earlier in the week, the federal agency opened a civil rights probe into the shooting, and in Florida, Seminole County State Attorney Norm Wolfinger said a grand jury will meet April 10 to consider evidence in the case.

"We are pleased the Department of Justice has heeded our calls and agreed to investigate this outrageous case," Jealous said in an emailed statement Tuesday. "The rules of justice in this nation have failed when an innocent teenage boy can be shot to death by a vigilante and no arrest is made for weeks."

At the town hall meeting, more than 350 people packed into the Allen Chapel AME Church, which is located in a traditionally black neighborhood of Sanford. People jumped to their feet and cheered when local NAACP leader Turner Clayton Jr. said the federal Justice Department should not only review the investigation but also take over the Sanford Police Department.

Other civil rights leaders said the city's police chief should step down.

"This is just the beginning of what is taking place," Clayton said. "We're going to make sure justice prevails."

When The Associated Press tried to reach the police department Tuesday evening for comment, a dispatcher told a reporter to call in the morning.

Earlier Tuesday, an attorney for Martin's family revealed the teenager told his girlfriend just moments before he was killed that he was being followed.

"'Oh he's right behind me, he's right behind me again,'" 17-year-old Trayvon Martin told his girlfriend on his cellphone, attorney Benjamin Crump said.

The girl later heard Martin say, "Why are you following me?" Another man asked, "What are you doing around here?'" Crump said.

Crump told reporters Tuesday Martin cried out when a man bearing a 9mm handgun came at him. Police said Zimmerman, who was found bleeding from his nose and the back of his head, told authorities he yelled out for help before shooting Martin.

"She absolutely blows Zimmerman's absurd self-defense claim out of the water," Crump said of Martin's girlfriend, whose name was withheld.

Martin, who was in town from Miami to visit his father in Sanford, called his 16-year-old girlfriend in Miami several times on Feb. 26, including just before the shooting, Crump said. The discovery of the lengthy conversations, including one moments before the shooting, was made over the weekend by Martin's father, who checked his son's cell phone log, Crump said.

The teenager told the girl on his way back from the store he'd taken shelter from the rain briefly at an apartment building in his father's gated community, Crump said. Martin then told her he was being followed and would try to lose the person, Crump said.

"She says: 'Run.' He says, 'I'm not going to run, I'm just going to walk fast,'" Crump said, quoting the girl.

After Martin encountered Zimmerman, the girl thought she heard a scuffle "because his voice changes like something interrupted his speech," Crump said. The phone call ended before the girl heard gunshots.

The last call was at 7:12 p.m. Police arrived at 7:17 p.m. to find Martin lying face down on the ground.

Zimmerman was handcuffed after police arrived and taken into custody for questioning, but was released by police without being charged. Police have interviewed Zimmerman twice since then.

Crump called the treatment patently unfair and asked if Martin would have received the same treatment if he had been the shooter.

"We will not rest until he is arrested. The more time that passes, this is going to be swept under the rug," Crump said.

Crump said he plans to turn over information about the call to federal investigators; a grand jury in Seminole County is also likely to subpoena the records. The Florida Department of Law Enforcement is also involved in the state case.

Former federal prosecutors said there are limitations to a Justice Department civil rights probe, which typically would involve a sworn law enforcement officer accused of abusing his authority.

In this case, they said, it's not clear whether Zimmerman had any actual law enforcement authority or if the Sanford Police Department did anything improper. Zimmerman had a permit to carry a gun, but it was not required for his neighborhood watch patrol.

"I think the community has the feeling that there's some type of cover-up," said Jeffrey Sloman, former U.S. attorney in Miami. "At least the department's involvement makes sure it gets some review. He wasn't a police officer. I'm sure that this is going to be a tough case to prosecute."

Authorities may be hamstrung by a state "Stand Your Ground" law that allows people to defend themselves with deadly force and does not require a retreat in the face of danger. Asked Tuesday if that law needs change, Republican Gov. Rick Scott said "it's always positive to go back and think about existing laws."

During the town hall meeting in Sanford, Florida Rep. Geraldine Thompson promised the law's repeal would be a top priority for the state legislature's black caucus.

"If vigilante justice becomes the norm, will visitors feel comfortable coming to our state?" she said.

An online petition urging local authorities to prosecute Zimmerman had drawn more than 700,000 signatures at website Change.org as of early Wednesday. About 50 defense attorneys and protesters filled the lobby in the governor's office Tuesday to deliver a letter seeking an independent investigation and a task force to study racial profiling. They applauded when Scott came out of his office to talk to them.

"I will make sure justice prevails," Scott said. "I'm very comfortable that (state law enforcement) is going to do the right thing. They're not going to let somebody do something wrong and get away with it."
